Alpha thalassaemia is an inherited blood disorder caused by deletions or mutations in the alpha-globin genes (HBA1 and HBA2) on chromosome 16. This multiplex PCR test detects the most common large deletions that remove one or both alpha-globin genes, accounting for the majority of alpha thalassaemia cases worldwide.

Who Should Consider Alpha Thalassaemia Deletion Testing?

Individuals of Southeast Asian, Chinese, Mediterranean, Middle Eastern, or African descent with microcytic anaemia not explained by iron deficiency. Those with a raised HbH inclusion body test or abnormal haemoglobin electrophoresis. Couples planning a family where one or both partners are from high-prevalence populations, to assess the risk of Haemoglobin Bart hydrops fetalis (a lethal condition caused by deletion of all four alpha-globin genes).

Alpha thalassaemia carrier states (one or two gene deletions) are usually clinically silent or cause mild anaemia. HbH disease (three gene deletions) causes moderate anaemia requiring monitoring. Homozygous alpha-zero thalassaemia (four gene deletions) is incompatible with life. Carrier testing has important implications for reproductive planning.
